Abstract

In Egypt, due to the high rate of pollution, ocular infections are considered one of the main threatening problems that affect the Egyptian community. In order to encounter this problem, a microbiologic multi- district survey was conducted to detect the most prevalent causative agent of ocular infections in Egypt. Further, an antibiotic- sensitivity testing was applied to the isolated microorganism where Vancomycinwas found to be the most potent agent. The aim of the work in this thesis was to enhance local bioavailability of Vancomycin (VCM) by enhancing its ocular residence and possibly increase its corneal permeability. VCM- loaded SLNs were prepared by double emulsion (W/O/W) solvent evaporation technique. Two factorial designs were adopted to study the effect of different process and formulation variables as well as the effect of lipid on the formulated nanoparticles. The optimization of the results revealed that F6.3 is the optimum formula which was composed of GTP in lipid: drug ratio 1:1, 1% low molecular weight PVA and by applying 1 minute sonication time. In addition, DSC, FTIR, SEM and TEM were applied for further characterization of the optimized formula
