Abstract

The determination of related compounds is emerging as a very important issue in the field of pharmaceutical analysis. It requires highly specific and accurate methods of analysis to be able to resolve the interferences that may arise during the analysis of these compounds. Several related drugs belonging to different pharmacological and chemical groups were determined throughout the present study, namely, a mixture of glibenclamide and metformin HCl, bopindolol malonate, celiprolol HCl, 2-amino-4-chlorophenol (a potential impurity of chlorzoxazone), and atenolol. Two derivative spectrophotometric methods, including zero-crossing first derivative and ratio derivative spectrophotometry, were developed for the analysis of the antidiabetic mixture of glibenclamide and metformin hydrochloride in tablet preparations. Sensitive fluorometric methods were proposed for the determination of the β-blockers, bopindolol malonate and celiprolol hydrochloride, without interference of some co-formulated or co-administered drugs.
The method developed for bopindolol was successfully applied for content uniformity testing of its tablets while the method developed for celiprolol was applied for its determination in tablets and spiked biological fluids. A simple colorimetric method was proposed for the determination of 2-amino-4-chlorophenol as a potential impurity in chlorzoxazone bulk powder. Different experimental parameters were studied and the method was successfully applied for the analysis of the studied compound in spiked chlorzoxazone powder.
The β-blocker, atenolol, was also determined by a sensitive reversed-phase liquid chromatographic method with UV-detection without interference of some related drugs. Different chromatographic parameters were investigated and the method was applied for the determination of atenolol in different tablet preparations. The validation criteria of the developed methods were intensively studied. All the results were statistically analyzed and compared with those given by official or comparison methods.
