Abstract This study was carried out in tissue culture laboratory in Veg & Floriculture Department in the in vitro culture partition but the open field partition was carried out in the medicinal and aromatic nursery plants in horticulture Department during seasons of 2003/2004 – 2005/2006 For achieve this study basil seeds were used to in vitro germination on Murashige and skoog medium supplemented with micro and macro elements and other growth regulators for using the sterile explants in the other steps like multiplicate the number of explants and increasing the second metabolites as (essential oil percentage and main components percentage). •This study revealed the following results :- 1-The best commercial Clorox concentrations to decrease the contamination percentage was 40% for 20 minute. 2-Increasing number of shoots was by using BAP at concentration of 1 mg/L and using node with a pair of axillary buds as explant. 3-The highest number of roots for explant was obtained from media supplemented with NAA at concentration of 0.5 mg/L. 4- The mixture of beat moss and vermiculite and loamy soil gave the highest value for survival percentage and shoot productions. 5- Using BA at concentration of 5 mg/L gave the best result for essential oil percentage also using sucrose as a carbon source gave the highest value for essential oil percentage and main components. |
