Abstract The study aimed to evaluate impact of lnc-NEAT1 gene expression on drug resistance in CLL and found out the correlation between lncRNA - NEAT1 and multiple drug resistance genes (ATP-binding cassette (ABC) transporters) in CLL patients. It also correlated lncRNA-NEAT1 expression patterns with prognosis to outline its possible role in CLL outcome. A total of 40 CLL patients were selected to participate in the study and 10 healthy patients as a control group. NEAT1 expression was significantly downregulated in CLL patients compared to healthy control while, ABCA3 was significantly upregulated in CLL patients compared to healthy control. NEAT1 showed a statistically significant correlation with parameters Rai-staging, tumor differentiation status, histopathology of lymphocytes, CD38 and treatment response. However, there was no significant correlation with parameters age, gender, number of nodel, IGHV mutational status and ZAP-70. ABCA3 showed a statistically high significant correlation with parameters Rai-staging, tumor differentiation status, histopathology of lymphocytes, CD38, IGHV mutational status, ZAP-70 and treatment response. However, there was no significant correlation with parameters number of nodel, age and gender. In addition, we conduct ROC analysis to detect the potential application of NEAT1 and ABCA3 in the diagnosis of CLL. Our results revealed that the mRNA ABCA3 expression obtains an area under the curve (AUC) is 0.980 with 95% confidence interval: in contrast to 0.716 shown in lncRNA NEAT1 expression. In addition, the area under the curve is significantly different from 0.5 since p- value is 0.000 meaning that the logistic regression classifies the group significantly better than by chance. This means that mRNA ABCA3 is good predictor marker for CLL with 98.0% sensitivity and 90 % specificity. On the other hand, lncRNA NEAT1 gene expression is not predictor for CLL. |
