Abstract

This study aims to investigate the effect of hibiscus (Hibiscus sabdariffa) and turmeric (Curcuma longa) for cyclosporine injected in male albino rats .
Forty two white male albino rats, Sprage Dawely strain, 2- week age, weighting ( 110-140 g ) will be used.
The rats will be acclimatized for a week under laboratory conditions before commencement of the experiments .
Cyclosporine will be used as single intraperitoneal injection (50 mg/kg/day) subcutaneous in olive oil for 10 days .
This group was further divided into the following subgroup :
• group (1): (Control ” – ”) rats (n=6) normal rats feed on basal diet only.
• group (2): (Control ” + ”) rats (n=6) treated with cyclosporine then will fed on basal diet only.
• group (3): rats (n=6) treated with cyclosporine then feed on basal diet containing  2.5 % of hibiscus powder.
• group (4): rats (n=6) treated with cyclosporine then feed on basal diet containing  5 % of hibiscus powder.
• group (5): rats (n=6) treated with cyclosporine then feed on basal diet containing  0.5 % of turmeric powder.
• group (6): rats (n=6) treated with cyclosporine then feed on basal diet containing  1 % of turmeric powder.
• group (7): rats (n=6) treated with cyclosporine then feed on basal diet containing 3% mixture of hibiscus and turmeric.
At the end of the experimental ( 4 weeks ), the blood samples were collected after12 hours fasting from the portal vein; the rats were scarified under ether anesthetized, blood samples were received into clean dry centrifuge tubes and left to clot at room temperature, then centrifuged for 10 minutes at 3000 r.p.m to separate the serum. serum was carefully aspirated and transferred into clean cuvette tubes and stored frozen at -20°C for analysis.
Next to the other, the organs heart, liver, kidney, spleen and lung were removed washed in saline solution, dried by filter paper and weighted. Biological and Biochemical changes were investigated. Liver and kidney samples were directly preserved and fixed in 10% neutral buffered formalin. For histopathology examination.
The data were statistically analyzed using a computerized SPSS program by one way ANOVA for a completely randomized design using a statistical analysis system. The results are presented as Mean +SD differences between treatments at P<0.05 were considered significant.
The results arrived at revealed the following:
1. The incidence of suppress immunity reduced the body weight gain, feed intake as well as feed efficiency ratio and it was gained by feeding the groups with hibiscus and turmeric powder, the best group was recorded for group (3) which contained hibiscus powder 2.5%.
2. The relative weights of the internal organs (liver, kidney and heart) of diabetic rats increased compared to the negative group, but the groups that were fed hibiscus and turmeric powder decreased the enlargement of the internal organs and their relative weights decreased compared to the negative control group in the rats. the best treatment show a decrease in the maximum relative weights of rats recorded by the group(6) which contained turmeric powder 1%.
3. Suppress immunity has led to a significant increase in the levels of liver enzymes. These rates were reduced by feeding the groups on hibiscus and turmeric powder. The levels of liver enzymes in group (6) which contained turmeric powder 1% were very close to the control group.
4. Suppress immunity leads to an increase in blood urea, creatinine, uric acid, total protein, albumin and globulin levels, while it decreases when hypercholesterolemia rats are fed hibiscus and turmeric powder. group (4) which contained hibiscus powder 5% best results.
5. Suppress immunity led to a rise in the TNF while led to a low in immunoglobulin (IgA –IgG – IgM ) was reduced. On the other hand feeding rats on hibiscus and turmeric powder reversed these changes. the best group (6) which contained turmeric powder 1%.
6. Suppress immunity caused disorders in the serum glucose level leading to the increased, while rats fed with hibiscus and turmeric powder recorded an improvement in it especially the group(4) that was fed hibiscus powder 5%.
7. CAT and SOD decreased due to suppress immunity , On the other hand feeding rats on hibiscus and turmeric powder reversed these changes. the best group (6) which contained turmeric powder 1%.
8. Suppress immunity led to low in Hemoglobin, Red blood cells, Hematocrit and MCHC while led to rise in MCH and MCV . on the other hand fed on hibiscus and turmeric powder reversed these changes. The best group (6) which contained turmeric powder 1%.
