Abstract

Polycystic ovarian syndrome (PCOS) is caused by abnormal production of androgens, resulting in the formation of small fluid-filled sacs in the ovaries. This study aims to shed light on Nigella sativa and licorice extracts and their effects on polycystic ovaries. Seventy adult female Wistar albino rats, weighing 160±10 g, were randomly divided into seven groups (10 rats each) as follows: group (1): normal rats act as negative control groups, group (2): rats those were subjected to oral administration of Nigella stevia ethanolic extract (NEE) at a dose (150 mg/kg/day), group (3): rats those were subjected to oral administration of licorice aqueous extract (LAE) at a dose (100 mg/kg/day), group (4): rats those were subjected to injection intramuscularly of at a dose (0.2 mg/kg weekly) of Estradiol® as a positive control group, group (5): rats Estradiol®-induced polycystic ovary those were treated with oral of NEE (150 mg/kg/day), group (6): rats Estradiol®-induced polycystic ovary rats those were treated with oral of LAE (100 mg/kg/day) and group (7): rats Estradiol®-induced polycystic ovary those were treated with oral of (50 mg/kg/day) reference medicen (Glucophge®) for six weeks. The results showed that Nigella sativa and Licorice powders contain moisture, ash, protein, crude fiber, crude fat and carbohydrates (15.24 ,4.93%), (0.78, 9.13 %), (13.56 , 8.62 %), (2.1 , 46.08 %), (0.97 , 2.62%) and (67.35 , 71.38%); respectively. It also contains calcium, sodium and phosphorus (571.5, 191.1) , (22.88, 3.64) and (704.5, 277.5)mg /100g respectively. Nigella sativa and licorice also contain 16 phenolic compounds. The results also showed that Nigella sativa and licorice extract succeeded in reducing ovarian cysts. This was demonstrated by a significant decrease in serum follicle-stimulating hormone, luteinizing hormone, prolactin, estradiol and testesterone, and an increase in progesterone and insulin. The results also showed a significant decrease in liver enzymes, urea, creatinine, total cholesterol, triglycerides, LDL cholesterol, glucose, potassium, MDA, NO, GSH, TNF-α, IL1β, IL-4, IL-6 and IL-10, in addition to the significant improvement in albumin, total protein, HDL cholesterol, calcium, sodium, SOD, GPx, CAT, and CD4. The study also showed the occurrence of histological changes in the ovarian tissue of experimental rats. Therefore, the study recommended the use of Nigella sativa and licorice in food because of their many benefits.
