Abstract

The use of dietary antioxidants to modulate the toxic side effects induced by the anticancer drugs used in chemotherapy is currently eliciting considerable interest. This study was undertaken to investigate the possible protective role of the antioxidant rosemary (Rosmarinus officinalis L., ROS) against oxidative stress induced by the anticancer drug Docetaxel (DTX) in a mice model with Ehrlich solid carcinoma (EAC). As Solid lipid Nanocarriers have been reported to efficiently improve pharmacokinetic profile, targeted drug delivery to tumor sites and release drugs with proper control, rosemary leaves extract loaded onto Solid lipid Nano-carriers are used in this study to protect it from gastrointestinal degradation and raise its therapeutic efficacy. Tumor-bearing mice were prepared by injecting the left flank of mice with 2 x 106 viable Ehrlich Ascites Carcinoma (EAC) cells / mouse in a volume of 0.2 ml. The tumor was allowed to grow for approximately 15 days then mice were haphazardly distributed to 7 different groups (10 animals / group) plus normal control group (8 total groups). The 1st group, control ”Normal control” that was not induced with tumor and received distilled water only. The 2nd group, Positive control (EAC) injected with Ehrlich ascites carcinoma and received distilled water alone. The 3rd group, (EAC) treated with Solid lipid nanoparticle (SLN). The 4th group, (EAC) treated with Docetaxel alone (DTX). The 5th group, (EAC) treated with rosemary loaded SLN (200mg /kg). The 6th group, (EAC) treated with rosemary loaded SLN (400mg /kg). The 7th group, received combined treatment of DTX) + (ROS) 200. The 8th group received combined treatment of (DTX) + (ROS) 400. At the termination of the treatment, liver samples were collected for performing oxidative stress biomarkers, biochemical parameters and liver function tests (serum ALT & AST). Mortality rate, Tumor size, Tumor Necrosis Factor-alpha (TNF-α) and hematological parameters were also estimated. In addition, Histopathological lesions in liver tissues and flow cytometric analysis were also investigated. Our results indicated that treatment with rosemary loaded SLN alone or combined with docetaxel showed improvement in all tested parameters as compared to group treated with DTX alone. In conclusion, Rosemary loaded SLN shows promising anticancer efficacy due to its antioxidant activity.
