Abstract

Isoxicam is a member of the oxicam family that inhibits both types of cyclo-oxygenase enzyme Cox-1 and 2. Fertility in females is affected clearly by COX-2. So, there is an association between use of isoxicam and miscarriage.
Melatonin is one of the antioxidants that prevent lipid peroxidation and formation of free radicals that leads to destruction of tissue. The aim of the study was to demonstrate the injurious effects isoxicam on the ovary of adult albino rat and to evaluate the protective role of melatonin in preventing the hazardous effects on the ovary.
Fifty-four adult albino rats separated into three groups. Each group consisted of eighteen rats.
group I (control): rats injected with normal saline(0.9mg/L).
group II (isoxicam): rats injected with 0.7gm/Kg Isoxicam intraperitoneally for 20 days.
group III (Isoxicam and melatonin): rats injected with 0.7mg/Kg Isoxicam + 10mg/kg melatonin intraperitoneally for 20 days. The rats were sacrificed, and their ovaries were dissected. Ovaries were examined by the light microscope and the electron microscope. The study revealed that isoxicam caused histological changes represented in degeneration of germinal epithelium layer and destruction of the ovarian follicles. In the group (III), receiving isoxicam and melatonin, there was restoration of normal architecture of the ovary tissues. There was an intact ovarian surface with some healthy ovarian follicles. These results are similar to the control as melatonin acts as antioxidant that prevents formation of free radicals that destroy tissue.
