Abstract

Tumor necrosis factor-alpha (TNF-Ü) is an important cytokine in generating an immune response against infection with hepatitis C virus (HCV). In HCV infection, the production of inappropriate cytokine levels may contribute to viral persistence. The G {uF0E0} A transition at position -308 and -238 in the TNF-Ü promoter has shown to affect the overall expression and the secretion of TNF-Ü. The aim of this study was to investigate the prevalence of genotypes associated with polymorphisms of TNF-Ü promoter at position -308 and -238 in a group of Egyptian HCV- infected patients compared with healthy uninfected control subjects. In addition, the association between these polymorphisms and the clinical characteristics of the positive HCV patients were investigated. Genotyping was determined by polymerase chain reaction (PCR) followed by restriction fragment length polymorphism analysis (RFLP) on genomic DNA isolated from 61 HCV patients and 20 healthy uninfected control
