Abstract These studies were conducted during the period from 2008 to 2014 at Department of Genetics, Faculty of Agriculture, Cairo University, Giza, Egypt, to improve tomato tolerance to salinity using biotechnological techniques. Development of transgenic tomato plants harboring glutaredoxin- 2 gene (GRX-2) was aimed. In the first experiment, a regeneration system has been established for five tomato cvs, i.e. Flora-Dade, Marmande, Summer Prolific, Castlerock and Super Strain B. Cotyledon and hypocotyl explants from the five tomato cvs were cultured on MS medium supplemented with different concentrations of plant growth regulators. Data showed that, cultivars and BA concentration significantly affected callus induction, shoot induction, and regeneration frequencies for each type of explants. The best cultivar was 2Castlerock3 at 6 mg/l BA.
