Abstract

Background:Influenza A virus is a negative sense single stranded enveloped RNA virus belong to family orthomyxoviradae. Flu virus causes annual epidemics and occasional pandemics. Annual outbreaks of influenza infection are an ongoing public health threat, and kill an estimated 250,000-500,000 people each year worldwide. Materials and Methods: The purpose of this study is developing new antiviral therapeutics against influenza viral entry and Identification of the host proteins involved in influenza viral entry. The study included 20,000 small molecule compound and 21,000 host genes using tissue culture, plaquing assay, western blott, Flowcytometry, fluorescence microscopy and robotics. Results: By Highthroughput screening by robotics we discovered a completely new two small molecule compound inhibitors that inhibit influenza virus at micromolar and nanomolar concentrations. They inhibit entry step at the fusion level. We discovered also a new host gene SLC23A2 that helps influenza virus infection and by knocking down this gene it inhibit influenza entry by 40 and 60% respectively
