Abstract

Background: It is estimated that approximately 10% of the population suffer a single convulsive episode during their lifetime. Epilepsy is the second most common chronic neurological disorder after stroke affecting approximately 0.5% of the population in developed countries and 1.5-2% in developing countries, especially male in developing countries, including African areas. Isoniazid (INH), a known first-line anti-tuberculous drug is claimed to cause imbalance in both GABA and glutamate brain levels, in favor of glutamate. Pentylenetetrazole (PTZ) is a well-established systemic chemoconvulsant that acts by blocking the GABAergic system. Also, kindling model is considered a chronic animal model of human complex partial and temporal lobe epilepsy. Aim: The purpose of this study is to explore the role of the phosphodiesterase-4 inhibitor (PDE4I), rolipram and the angiotensin converting enzyme inhibitor (ACEI), ramipril by comparison to the benzodiazepine (BZ) agonist, diazepam, in isoniazid-induced seizures and PTZ kindling model of epilepsy in rats
