Abstract

Aim of the study: To assess the effect of Odanacatib as a Cathepsin K enzyme inhibitor on Orthodontic tooth movement in rabbits. Materials and methods: Eighteen male 18 months old New Zealand white rabbits were divided equally into two groups and then fitted with a NiTi coil spring extending from the mandibular right 1st premolar to the mandibular incisors, creating a mesializing force on the 1st premolar. The test group received 5mg/kg ODN intraligamentary injection on days 1, 7 and 14 of the 3-week experiment. The control group followed the same protocol as the test group, but with 0.9% NaCl solution instead of ODN. Results: The rabbits in the test group showed reduced tooth movement than the control group. The spaces created distal to the mandibular right first premolar were greater with a mean distance of 1.19 ± 0.07mm in the control group comparing to a mean of 0.53 ± 0.08mm in the intervention group. The results were statistically significant, where p<0.05. Conclusions: from the findings of the current study, the following conclusions could be drawn; local administration of Odanacatib was found to have a statistically significant retarding effect on the orthodontic tooth movement in rabbits in comparison to the control group. Bone resorption at the compression sites of teeth in the tested group was less than the control group indicating decreased osteoclastic activity. The fractal dimension analysis of both groups showed that the ODN-treated group exhibited less irregularity in the PDL-bone interface contrary to the control group. Further research is required to investigate the application of this drug on humans for the same objectives of this study and to weigh the benefit/ risk ratio profile of ODN
