Abstract

The efficacy of filgrastim and Ü-tocopherol succinate (Ü-TCS), alone or combined was investigated in gastrointestinal acute radiation syndrome (GI-ARS) induced following 10 Gray whole-body Þ-irradiation. Mice were randomly allocated into 10 groups.The first group received saline and was not exposed to {u0263} rays and served as normal control. Groups 2-10 were all exposed to {u0263} rays (10 Gy). group 2 received saline and served as irradiated control. Groups 3 & 4 were treated with filgrastim 5 æg/Kg & 10 æg/Kg respectively starting 1 hour after irradiation daily for four successive days. Groups 5 & 6 received a single dose of 200 mg/kg & 400 mg/kg Ü-tocopherol succinate (Ü-TCS) 24 hours before being exposed to the {u0263} rays respectively. Groups 7 & 8 received combined doses of the small dose of filgrastim with the low dose of Ü-TCS and the high dose of filgrastim with the low dose of Ü-TCS respectively. The last two groups received the low dose of filgrastim with the high dose of Ü-TCS and the high dose of filgrastim with the high dose of Ü-TCS respectively. Treatments with filgrastim in all assigned groups were started 1 hour after irradiation daily for four successive days, while, treatments with Ü-TCS in all assigned groups were given once 24 hours before irradiation.Serum citrulline level, intestinal interleukin-1Ý (IL-1Ý), reduced glutathione (GSH) and malondialdehyde (MDA) contents as well as myeloperoxidase (MPO) activity were measured. Intestinal caspase-3, p53, cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) immunopositivity were examined
