Abstract The purpose of this study was to examine the effect of cisplatin on histological structure of cerebellum and possible protective role of granulocyte colony stimulating factor (G-CSF) versus citrullus lanatus juice. Forty adult male albino rats were divided into control group, group I: rats received cisplatin by intraperitoneal (IP) injection at a dose of 10 mg/kg on days 6 and 13, group II: rats received G-CSF by subcutaneous (SC) injection at a dose of 50 æg /kg for 5 consecutive days then cisplatin on days 6 and 13 and group III: rats received citrullus lanatus juice daily (for 16 days) by oral gavage at a dose of 4ml/kg in concomitant with cisplatin on days 6 and 13. Blood samples were taken to measure plasma malondialdehyde (MDA) and reduced glutathione (GSH) levels. Cerebellar biopsies were processed for light microscopy using H&E, Bielschowsky silver and immunohistochemical stains (glial fibrillary acidic protein (GFAP) & caspase-3) as well as for electron microscopy. Morphometric and statistical studies were done for mean number of Purkinje cells, mean thickness of granular layer, mean area percent of GFAP positive cells and mean optical density of caspase-3 positive cells |
