Abstract This Thesis includes four chapters. The first chapter embodies an introduction about Gewald reaction and its modifications, also the recent trends in thieno[2,3-d]pyrimidine derivatives, as well as a description of their anticancer activities. The second one includes the research objectives and Schemes for the synthesis of the recent target compounds. The third one illustrates the discussion of the experimental work of the starting compounds I and VI also intermediates II, III, VII, VIII, IX, XI and the new target compounds IVa-c, Va-k, Xa-f and XIIa-i, as well as spectroscopic confirmation of the intermediates and the newly synthesized target compounds. Furthermore, this chapter comprises the discussion of the anticancer activity of the new target compounds and their molecular modeling study. The fourth chapter consists of the experimental work of the preparation of all the synthesized compounds conforming to three Schemes. Scheme 1 implicates the synthesis of starting compound 2-amino-5,6,7,8-tetrahydro-4H-cyclohepta[b]thiophene-3-carbonitrile (I) followed by its reaction with formic acid to give II that was reacted with phosphorus oxychloride to afford III.
