Abstract The present study was conducted to investigate changes in expression of serum XO enzyme during hyperuricemia and the ameliorating effect of natural safe source plants on the hyperuricemia in rats. For this purpose, 50 male albino rats were categorized into 5 equal groups as follows: (GI) Control group, (GII) Hyperuricemic group, (GIII) Febuxostat Hyperuricemic group, (GIV) G. biloba hyperuricemic group, (GV) Onion juice hyperuricemic group. At the end of 4 weeks, blood samples were collected and sera were separated and analyzed for determination of serum XO, ALT, AST, ALP activities, UA , urea, creatinine and TP. conc. , liver and kidney specimens divided into 3 parts , Part for estimation of MDA, GSH, uric acid contents and SOD, GSH-Px activity , Part for histopathological examination and Part (liver only) for studying XO gene expression The obtained results were statistically analyzed and the general findings can be summarized as follows: PO caused hyperuricemia and increment in XO gene expression (slightly), serum XO, uric acid in serum and tissue, urea and creatinine, caused various degrees of pathological damages in liver and kidney. Febuxostat decrease XO gene expression, serum XO, uric acid in serum and tissue, urea, creatinine, both GBE and onion juice decrease XO gene expression (slightly), serum xo, UA in serum and tissue, creatinine and relieved PO pathological changes in liver and kidney. We conclusion, our findings suggested that the two studied plants (Gingko biloba and Onion) can be considered as a source of natural antioxidants and xanthine oxidase inhibitors, that can be developed, upon further investigation, into successful herbal drugs for therapeutic application against gout and hyperuricemia. |
