Abstract

Purpose: Gentamicin is an aminoglycoside antibiotic widely used against Gram-negative microorganisms. Nephrotoxicity is the main limitation to its therapeutic efficacy.The aim of work was to investigate the role of dried and germinated fenugreek seeds as a potent antioxidant against the nephrotoxicity and oxidative stress in the kidney tissues. Materials and Methods: 36 Male Swiss albino mice were divided into 6 groups (6 mice in each group) group I: the animals were injected intraperitoneally with distilled water. group 2: the animals were intraperitoneally injected with gentamicin. group 3: the animals were orally received fenugreek (Trigonella foenum -graecum). group 4: the animals were orally received germinated fenugreek. group5: the animals were intraperitoneally injected with gentamicin simultaneously with fenugreek (Trigonella foenum -graecum). group 6: the animals were intraperitoneally injected with gentamicin simultaneously with germinated fenugreek. All the treatments of gentamicin were given as (100mg/kg) and all treatments of dried and germinated fenugreek were given as (500mg/kg) a single daily dose for five days. Physiological studies were applied on kidney by measuring kidney function parameters, oxidative stress parameters, as well as histopathological examination. Results: Administration of gentamicin caused blood urea, serum creatinine, uric acid; and Lipid peroxidase, (MDA) levels were raised significantly, while the reduction in levels of reduced glutathione, (GSH); glutathione reductase, (GR); superoxide dismutase, (SOD); and catalase, (CAT) as compared to control. Renal damage was confirmed by histopathological studies. Co-administration of dried and germinated fenugreek along with gentamicin led to significant restoration of blood urea, uric acid, serum creatinine MDA, GSH, GR, SOD, and CAT levels
