Abstract Radioimmunoassay is highly sensitive laboratory technique used to measure minute amounts of substances including antigens, hormones, and drugs present in the body. This vitro assay can calculates the occurrence of an antigen with very high sensitivity. Mainly, any biological product or substance for which a specific antibody exists can be measured, even in minute concentrations of picomolar and nanomolar concentrations of hormones in biological fluids. The preparation of high technology radioimmunoassay (RIA) reagents with low cost is considered the main objective of the present study to facilitate measurement of ferritin concentration in human serum as an essential parameter for investigating iron deficiency anemia and other diseases. The objective of the present work was: The extraction and purification of ferritin, production of ferritin polyclonal antibody, production of second antibody as separating agent, preparation of ferritin standards and radiolabeled ferritin tracer. The prepared system undergoes extensive optimization and validation studies. The produced RIA system proved to be specific, sensitive, precise and accurate. The system can be used for routine laboratory uses to detect serum ferritin concentration. |
