Abstract

Dopamine is the key to our happiness and reward feelings. It is a vital hormone and neurotransmitter for the human and animal body. Dopamine functions are responsible for controlling mood, memory, movement, body weight, learning, cognition, attention, sleeping, and other processes that affected the quality of life. It is synthesized only in the brain, then spreads throughout the entire body as signals by binding to its receptors especially dopamine receptor D3 (DRD3) the most important one. Phenylalanine (Phe) is the precursor for dopamine, and phenylalanine hydroxylase enzyme (PAH) is the control valve to start the biosynthesis of the dopamine process. Many types of diet contain phenylalanine, but faba bean Sakha3 (genotype) is one of the highest sources of phenylalanine. In the present study, male New Zeland rabbits were used as an ideal model for measuring the effect of a diet rich in (high phenolic acid) like faba bean (dry and fresh forms) of Sakha3 genotype on the gene expression, protein expression of phenylalanine hydroxylase enzyme (PAH) in rabbits (isolated from the liver of rabbits) and on the gene expression of DRD3 (isolated from the brain of rabbits).Results confirmed that the diet as the dry form of Sakha 3 (genotype), increased the levels of phenylalanine more than three times in blood samples of rabbits that were fed to it, which were measured using the phenylalanine assay kit. The dry form of Sakha3 also positively affected the gene expression, which was determined by using the (qRTPCR) technique and the protein expression of phenylalanine hydroxylase, which was determined by using the Western blotting technique. The results were confirmed significantly increased in the gene expression of PAH in rabbit was fed on dry form of Sakah3, It increased three times more compared to the gene expression seen in the control rabbit. Also, the protein expression in the same rabbit showed the highest gene expression compared to the gene expression of rabbit was fed on fresh form of Sakha3 and control rabbit.
