Abstract

Human toxoplasmosis is a widely distributed infection which cause serious congenital sequelae and fatal illness in immunocompromised, recently studies also showed its effect on mental health and personality of immunocompetent patients so efficient diagnosis of toxoplasmosis is essential. Merging the innovative use of Quantum dots and the widely used immunoassays can lead to a better diagnostic performance. The study included175 individuals divided after examination using Sabin Feldman dye test (SFDT) which is considered the gold standard test for diagnosis of toxoplasmosis into group 1a: Symptomatic patients with sera positive for Toxoplasma antibodies with titers <1:1024, group 1b: Symptomatic patients with serum positive for Toxoplasma antibodies with titers ≥ 1:1024 suggestive for acute infection(IgM antibodies), group 2: Symptomatic patients with serum samples negative for Toxoplasma antibodies by SFDT, Group3: Non symptomatic patients, negative for Toxoplasma antibodies and positive for other parasitic diseases and group 4: Non symptomatic apparently healthy control. Cadmium selenide/ Zinc Sulfide (CdSe/ZnS) nanobeads was conjugated to IgG/ IgM anti-human horseradish peroxidase conjugate for detection of T. gondii IgM/ IgG antibodies. the sensitivity, specificity, positive predictive value, negative predictive value and relative agreement of Quantum dots nanobeads based immunoassay in detection of IgG Toxoplasma antibodies in relation to indirect ELISA was 87.4%, 90.9%, 90.5%, 87.9% and 89.1% respectively. And in IgM Toxoplasma antibodies detection was 81.0%, 95.7%, 85.0%, 94.4% and 92.3% respectively. It was concluded that QDNBs based immunoassay improves the performance of ELISA particularly sensitivity as well as shorten the assay time.
