Abstract Donkeys’ popularity (Equus asinus) increased as working livestock in Egypt. The present study was carried out to investigate effect of ovarian function, in vitro maturation, sperm collection, sperm capacitation and in vitro fertilization.Pairs of Jennie’s ovaries were collected separately in plastic bags from slaughter Giza Animal Zoo slaughter house (2014 to 2016). The number of ovarian follicles was determined. A total of 234 oocytes were recovered from 148 Jennie’s ovaries. During the course of study, the number of follicle /ovary was calculated then oocytes were aspirated by syringe the number of oocyte/ovary was calculated. The recovered oocytes were classified according to the appearance of cumulus cells into excellent, good, fair, and poor. In this study three different maturation media were used for maturation of Jennies’ COCs TCM199 (n=5o), CR1aa (n=50) CR2aa (n=50), Follicular fluid with cells(n=50) and Follicular fliud without cells(n=50) for 36 h at 38.5ºC under 5% CO2 in humidified air. Maturation rate was assessed on the basis of Cumulus cells expansion and presence of the first polar body. The semen extender were SP- TALP, Equipro and kenney. Sperm capacitation by control, heparin, calcium ionophores, follicular fluid and hyaluronic acid. Assesment of fertilization rate and fertilization rate. The result obtained revealed that the Follicular fluid with cells was the best for in vitro maturation. Equipro the best sperm extender. The hyaluronic acid was recorded highly significant rate of jack sperm caacitation than other agent. Follicular fluid with 100% concentration revealed highly cleavage rate than other concentration. |
