Abstract
Boiling of the N-phenyl-2-[2-(quinolin-4-
ylamino)acetyl]hydrazinecarbothioamide (1) in 2N NaOH solu-tion at
refluxing temperature afforded 4-phenyl-5-[(quinolin-4-ylamino)methyl]-
4H-1,2,4-triazole-3-thiol (2) in 75% yield. Alkylation of the 1,2,4-triazole
thioles/thiones 2 with methyl or ethyl iodide in alkaline medium afforded
the corresponding N-{[(5-(methylthio)-4-phenyl-4H-1,2,4-triazol-3-
yl]methyl}quinolin-4-amine (3a) in 72% yield or N-{[5-(ethylthio)-4-
phenyl-4H-1,2,4-triazol-3-yl]methyl}quinolin-4-amine (3b) in 75% yield,
respectively. Hydrazinolysis of 3a and 3b gave N-[(5-hydrazinyl-4-
phenyl-4H-1,2,4-triazol-3-yl)methyl]quinolin-4-amine (4) in 78% yield
(Scheme 1).
