Abstract

Exposure of the body to ibuprofen produces reactive oxygen species (ROS) which damage proteins, lipids and nucleic acids. Because of the lipid component in the cell membrane, lipid peroxidation is reported to be susceptible to ibuprofen nephrotoxicity. So the present study was performed to detect the role of Olive and Ocimium bacilicum leaves extracts (100 mg/kg b.wt.) to overcome the side effects of ibuprofen. The parameters studied in the current work were serum Urea, Creatinine, Uric acid, erythropoietin, homocysteine, interleukin 1beta, interleukin 10, ALT, AST, proteins profile (total protein, albumin), lactate dehydrogenase and creatine kinase, liver and kidney lipid peroxidation, catalase, superoxide dismutase (SOD) and glutathione (GSH) Content.
Male albino rats (36) were used, weighing 110-140g, divided into 6 groups, each consists of 6 rats:
group 1 was normal control group, group 2 consisted of rats administrated single dose of ibuprofen (15mg/kg body weight/ day), group 3 received ethanolic olive leaves extract, group 4 received ethanolic extract of basil leaves, group 5 received ibuprofen followed by ethanolic extract of olive leave, group 6 received ibuprofen followed by ethanolic extract of basil . Olive leaves extract and basil leaves extract were emulsified with saline in a dose of (100 mg/ kg body weight / day) for 4 weeks.
Sacrifice of all animals was performed at the end of the experiment and blood, kidney and liver were obtained for determination of different biochemical parameters.
The results of the present study can be summarized as follows:
110
Summery
1- Rats administrated ibuprofen exhibited a profound elevation of urea, creatinine, uric acid, homocysteine, interleukin 1beta, alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase and creatine kinase abnormalities and an increase in lipid peroxidation level. Noticeable DROP in liver and kidney catalase, superoxide dismutase, glutathione content, serum erthropietin, interleukin 10 and total protein, albumin levels were also recorded.
2- Rats treated with either olive leaves extract or basil leaves extract showed significant modulation in urea, creatinine, uric acid, homocysteine, transaminases, interleukin 1beta and noticeable improvement in the activity of antioxidants (catalase, superoxide dismutase and glutathione) and protein profile (total protein, albumin), erythropoietin and interleukin 10 . Olive and basil leaves extract was also effective in minimizing the ibuprofen-induced increase in lipid peroxidation in some tissue organs comparing with control rats.
