Abstract

Background: Hepatocellular carcinoma is considered a global problem, as it represents the sixth most common cancer worldwide. Since gene expression disorders are the most common causes of cancer development. There had been a noticeable interest in studying these disorders for multiple genes in order to use them as tools for early HCC diagnosis and patients follow-up. The current study studied the genetic expression of laminin beta 1 and keratin 19 in order to ensure that they can be used as new biomarkers of liver cancer.Objectives: Evaluation of the potential predictive value of both laminin beta 1 and keratin 19 in early diagnosis of liver cancer.Methods: 3 ml of blood were withdrawn from all the subjects included the study. The WBCs were deposited as a pellet by using the RBCs lysis buffer. Extraction of the RNA from the WBCs pellet. Reverse transcription of the extracted RNA in order to form cDNA. The quantitative real time PCR reaction was done to quantify the mRNA expression of laminin beta 1 and keratin 19.Results: The results of the current study showed that keratin 19 is characterized by a high ability to differentiate between the HCC group and the cirrhotic control group as well as the healthy control group. The study also showed that keratin 19 had a higher ability to early diagnose hepatocellular carcinoma than laminin-beta 1.Conclusion: from the current study it can be concluded that keratin 19 can be considered as a new biomarker for early diagnosis of hepatocellular carcinoma.Keywords: Hepatocellular carcinoma, Laminin beta 1, Keratin19.
