Abstract

Poultry industry is the most popular emerging industry in the world as it supplies people with premium quality of animal proteins i.e. meat and egg that alleviates the huge protein gap and malnutrition problem for the people around the world. This study was carried out to detect genetic polymorphisms in three broiler breeds (Avian 48, Cobb 500 and Ross 308) using RAPD-PCR, TLR4 and IGF-I genes polymorphisms using (PCR-RFLP), genetic expression of (TLR4 and IGF-I) using quantitative real time PCR and serum analysis of (IL-10 and IGF-I) by ELIZA test. Blood samples from one hundred chicks from each breed were collected into EDTA containing vacutainer tubes to extract DNA for (RAPD and RFLP) while blood samples for serum analysis were collected without anticoagulant into a clean dry centrifuge tube. Ten samples from both liver and spleen from each breed were collected to extract RNA for gene expression. 42 bands were produced in RAPD. Out of these bands, 3 were recognized as monomorphic and 18 as polymorphic bands. The highest percentage (67%) of polymorphic bands was generated by OPC-1, while the lowest percentage (20%) of polymorphic bands was generated by OPA-20. Interbreed of band sharing frequencies were 0.82±0.13 between Avian-Cobb, 0.76±0.19 between Avian-Ross and 0.85±0.09 between Cobb-Ross. Three primers (OPA-8, OPA-10 and OPA-20) were specified for Ross, Avian and Cobb breeds, respectively. No polymorphism was detected in IGF-I gene using PstI enzyme among the three breeds. TaqI digestion of TLR4 gene gave BB genotype in the three breeds and AB genotype in Avian and Ross only. Also, the biochemical serum analysis of this study was compatible with gene expression analysis reflecting that Avian breed has higher immunity and growth rate than both Cobb and Ross breeds, respectively.
