Abstract All animal experiments were conducted in accordance with protocols approved by the Ohio State University Institutional Laboratory Animal Care and Use Committee. A total of 120 mice (weighing 18+ gm.) were used in this study. Adult female C57BL/6J WT and B6.Cg miR-155 KO mice were obtained from Jackson Laboratories. Mice were housed 3–5 per cage and provided standard food and filtered tap water ad libitum. Mice were anesthetized and surgery was performed under the microscope. A T9 laminectomy was performed to expose the dura. A moderately-severe contusion injury was induced using the IH device. The mice from both genotypes were distributed randomly into two main groups; group I: for PCR/ELISA and group II: for histology purposes. Each group was subdivided into naïve control subgroup and five subgroups according to time-point; 1, 3, 7, 14 and 42dpi groups. The experimenter was blinded to the groups during the surgery, animal care, tissue dissection, histopathology, imaging and data analysis. First experiment: fresh tissue samples at all time-points were collected and then used for performing PCR and ELISA. Second experiment: fixed tissue samples were used for histological analysis (WMS analysis, IHC). The locomotor recovery was evaluated with BMS before injury and on 1, 3, 7dpi then weekly up to 42dpi and were made by two blind observers. Force meter testing and gridwalk were conducted for mice from both strains at 35dpi for detection of muscle reflexes and evaluation of fine motor control respectively. |
