Abstract

Medications, especially (Acetaminophen) is widely used where it’s available on the table that doesn’t need a prescription to spend, it is analgesic Cytokines:Remarkable changes in biomarkers of immune acetaminophen treatment group were demonstrated by the substantial increase in the level of TNFα, IL-6, IL-1 β as compared to the control group. .Improvement was noteding.Therapeutic group given rutin, Intravenous injection of stem cells restord the level of cytokines the highest restorate was in the group treated with stem cells and rutin together Antioxidant redox indicators: Significant reduction in the level of glutathion (GSH), and Super oxide desmutase (SOD), with very high level in malondialdehyde (MDA) in liver tissue of rats treated with acetaminophen treatment compared with the control group On the other hand, improvement was noted in antioxidant levels for stem cell group with rutin followed by rutin group. where best results were obtained in The group given rutin and stem cells. Histopathological examination changes: Histological hepatic sections group rats given acetaminophen dilated central veins filled with inflammatory infiltrative cells and necrotic changes observed in some areas. While liver microscopy showed rats treated with stem cells rutin improvement in form and composition carcinomas lack the breadth of hepatic inflammatory cells also decreased .and antipyretic and has no side effects at the recommended dose limits, Scientific evidence indicates that the overdose and excessive use leads to the accumulation of the drug and cause liver toxicity in humans and animals. Herbal medicines have attracted a lot of attention lately as alternative medicines are useful for treatment and prevention of lifestyle disorders.(Rutin) is a natural antioxidant and also called gliykosid flavonoids and phenolic compounds are one of the many plants, which feature as anti-inflammatory, antioxidant, and anti-cancer.(Stem cells) are undifferentiated cells, That have the ability to regenerate unlimitedly and divide, and even switch to many other types of cells, The medium stem cells extracted from bone marrow have great potential in tissue repair and have the ability to configure the bone cells, fat, cartilage, muscles, nerve cells, liver cells, skin cells.This study was conducted on male rats of the number 120, medium weight (160-150), divided into five groups, each of which contains 10 rats as follows: 1. First group (control group): given distilled water only during the experiment. 2. Second group (acetaminophen): given the drug acetaminophen (750 mg/kg of body weight) every 72 hours for 21 days and left without treatment for 8 weeks. 3. Third group (group of acetaminophen with rutin): given the drug acetaminophen (750 mg/kg of body weight) every 72 hours for 21 days and legining from the third week given rutin(25 mg/kg body weight) per day for 8 weeks. 4. The fourth group (group of acetaminophen with stem cells): given the drug acetaminophen (750 mg/kg body weight) every 72 hours for 21 days and legining from the third week given stem cells injected intravenously in the tail and left for 8 weeks. 5. The fifth group (group of acetaminophen with rutin stem cells): given the drug acetaminophen (750 mg/kg of body weight) every 72 hours for 21 and legining from the third week given rutin (25 mg/kg body weight) per day and stem cells injected intravenously in the tail and left for 8 weekAfter 8 weeks animals were dissected and biochemical study included measuring liver enzymes Asparrtate amino transferase, alanine transferase AST, ALT, gamma-glutamyl amino transferase GGT, ALP and alpha fetoprotein AFP, cholesterol level, (TG, HDL, LDL) tumor necrosis factor (αTNF.), inter leukin-6 (IL-6), inter leukin -1beta (IL-1 β), cytochrom p450 (Cytochrom P450). Summary of the results obtained from the present study are follows:Biochemical changes-liver enzymes: The current study showed a marked increase in the level of liver enzymes in rats exposed to drug acetaminophen compared to the control group, Rutin and stem cell therapy group revealed while upper limit of improvement was in both rutin stem cells group and compared with the other groups.-Lipib profile level: for the acetaminophen group statistically significant rise occurred in cholesterol level, TG, LDL, while HDL level greatly reduced compared to the control group. Rutin and stem cells group recorded a partial improvement in the other treatment group.
