Abstract The present study was conducted at the Genetics and Genetic Engineering Lab, Faculty of Veterinary Medicine, Zagazig University. The main objective of this study was to detect polymorphisms in GDF9 and BMP15 genes in Barki and Rahmani sheep. In the present study blood samples from 100 ewes (50 for Barki and 50 for Rhmani) were collected from a private farm at Giza governorate. This study was aimed to determination of polymorphisms of GDF9 gene and BMP15 gene in Barki and Rahmani sheep by Polymerase Chain Reaction – Restriction Fragment Length Polymorphisms (PCR-RFLP), Single strand confirmation polymorphism (SSCP) and DNA Sequencing for detection of SNPs in the four exons of both breeds. Molecular genetic studies Polymorphism of GDF9 gene The PCR product of exon I of GDF9 gene was 712 bp, these amplicons had been treated with HhaI enzyme which cut the 712 bp into (120, 254 and 338) in all animals with no differences between all the tested animal of Barki and Rahmani sheep with absence of G1 mutation. Also SSCP result showed the same pattern revealing no differences in the sequence of this amplicon in both breeds. DNA sequencing had confirmed these results.
