Abstract

The present study was carried out to evaluate the possible adverse effects of doxycycline (20 mg/kg and 40 mg/kg) and fluconazole (2.5 mg/kg and 5 mg/kg) on total serum testosterone level, oxidative stress and antioxidant markers in adult cocks. Moreover histopathological examination of testes, epididymis, liver and kidney was done, with especial references to their effect on some hematological and biochemical parameters. A total of hundred and twenty five (125) adult cocks (1500 -1700 gm) were divided into five equal groups, (each of 25 cocks) as follow:1st group: cocks were served as non treated group (control).2nd group: cocks were given the therapeutic dose of doxycycline (20mg/kg b.wt) orally in drinking water for a 7 days. 3rd group: cocks were given the double therapeutic dose of doxycycline (40 mg/kg b.wt) orally in drinking water for 7 days. 4th group: cocks were given the therapeutic dose of fluconazole (2.5 mg/kg b.wt /day) orally in drinking water for 7 days. 5th group: cocks were given the double dose of fluconazole (5 mg /kg b.wt/day) orally in drinking water for 7 days.Blood samples were collected from 5 cocks of each group by slaughtering with sharp knife on 1 day and 1st, 2nd, 3rd and 4th weeks post drug administration. Samples were collected in two test tubes, First one mixed with salt of ethylene diamine tetra-acetic acid (EDTA) and subjected to heamatological examination. While other blood samples were collected in tubes (without anticoagulant) which put in centrifuge at 3000 /15 rpm to get a clear serum. The obtained sera were kept in deep freezer at – 20o C until assayed for serum total testosterone level and other biochemical paramrters. Effect of doxycycline and fluconazole on some hematological parameters were studied in clinically healthy mature cocks. The results showed that there was no significant variation in total erythrocytic count in doxycycline treated groups at both therapeutic and double therapeutic doses either between or in the same groups at different time. While Hb value was significantly increased at both concentrations of doxycycline in medicated groups. On the other hand, it was found that fluconazole at both doses (2.5 mg/kg and 5 mg/kg) showed a non significant changes in total erythrocytic count, while Hb value has showed a significant increase at both doses. The biochemical analysis of serum of clinically healthy mature cocks was carried out to estimate liver function tests (ALT and AST), kidney function tests (uric acid and creatinine), total antioxidant capacity, malondialdehyde and total testosterone hormone. The obtained data reflected a significant increase in serum transferases levels (ALT and AST) at therapeutic and double therapeutic doses of doxycycline medicated groups. Moreover, the recorded results revealed a significant increase in serum ALT level in the double therapeutic fluconazole treated group only on 1day and 1st week post dosing While serum AST recorded a non significant changes at both doses of fluconazole. The obtained results showed that doxycycline (20 mg/kg and 40 mg/kg) and fluconazole (2.5 mg/kg and 5 mg/kg) induced a significant increase in serum uric acid, creatinine, total antioxidant capacity and malondialdehyde levels. While there were a significant decrease in total serum testosterone hormone levels when compared to the results of control group. Histopathological examination of liver, kidney, testes and epididymis at 2nd and 4th weeks post treatment showed variable changes as a result of treatment with doxycycline (20 mg/kg and 40 mg/kg) and fluconazole (2.5 mg/kg and 5 mg/kg).
