Abstract

In this study we stand on the real cause of abortions, neurological signs, and still birth which spread among horse populations in many farms of Arabian horses in different areas in Egypt. We detect Equine herpes virus 1,2,4 and 5 in 202 field samples collected from horses stud had a history of cases of abortions in different stages of pregnancy, stillbirth, nervous signs, and respiratory affections. the collected samples were aborted feti, placenta, nasal swabs, blood, and cerebrospinal fluid. and the following steps was carried out:
1. SNT (serum neutralization test) used to detect EHVs in collected blood samples. After inactivations of serum sample the test was done and we find 110 tested serum samples, 59 and 78 were seropositive by SNT for EHV-1 and EHV-4.
2. Then we use the primer of EHV-1 to amplify a nucleotide sequences from the extracted DNA and also for EHV-2, EHV-4, and EHV-5.
3. Identification of PCR product using gel electrophoresis and check the PCR product by measuring with positive control.
4. There were 74 positive samples from total 202 tested samples by PCR against EHV-2 and 9 positive samples from a total 202 samples by PCR against EHV-4 as well as 83 positive samples from total 202 tested samples by PCR against EHV-1 and finally 43 samples from 202 against EHV-5.
5. In the phylogenetic study, partial sequences of gB and POL were used to conduct the analysis forEHV-1, EHV-2 or EHV-5 and to detect the relationship with other reference strains from GenBank. The data demonstrates that the obtained sequences are phylogenetically related to other EHV-1, EHV-2 and EHV-5 strains detected in different countries before.
6. Virus isolation was done using Equine dermal cell (ED) and rabbit kidney 13 (RK13).
