Abstract In a trial to investigate the recent situation of Newcastle disease virus at Sharkia Governorate among pigeons. Fifty one pigeon houses of both commercial and backyard rearing in the period between 2013-2015 were examined. The recorded clinical signs were nervous signs (torticollis, head tremors, leg paralysis and wing paralysis), greenish diarrhea, general signs of illness, emaciation and respiratory signs. On necropsy, congestion and hemorrhages were observed in brain, catarrhal enteritis with ulcers in the intestinal mucosa with greenish contents, nephritis and bronzy colored liver with or without necrosis were also recorded. The isolation of suspected viruses was tried in ECE. The results revealed embryo congestion and hemorrhage. Presence of haemagglutinating agent was determined using a rapid and plate HA assay of collected allantoic fluids in 41 samples (80.38%). The HI of the same viruses against NDV hyperimmune serum were positive in 38 virus isolates. While, the other three virus isolates failed to be inhibited by hyperimmune serum. The pathogenicity was carried out on the basis of MDT value in ECE revealed 72 ± 0.0 and 62.4±5.89 indicating that both isolates could be mesogenic strains. Allantoic fluid from each HA positive sample was subjected to RNA extraction followed by RT-PCR test using specific primers. |
