Abstract

In our study, although the expression of both FLT3 mRNA transcript and FLT3 receptor protein were higher among AML patients harboring the FLT3 ITD mutation, the results showed only a trend, however were not statistically significant. We determined a cut-off value for FLT3 receptor protein (CD135) of >49%, which would predict AML patients carrying FLT3 ITD mutation. Moreover, we found a statistically significant positive correlation between FLT3 mRNA and FLT3 receptor protein expression.
Finally, estimation of FLT3 receptor protein (CD135) expression by flow cytometry (more cost effective and less time consuming than other methods) in association with the routine leukemia immunophenotyping panel at the time of diagnosis of AML could be a good predictor for the FLT3 mutational status using a cut-off value of > 49%. Moreover, the frequent over-expression of FLT3 receptor protein in AML patients, with or without FLT3 gene mutations, points to its importance in selecting patients for whom FLT3 inhibitors therapy may be indicated.
