Abstract Mucositis is the inflammation of the oral mucosa following chemotherapy or radiotherapy in patients. The use of Ketoprofen as one of the anti-inflammatory agents is a promising strategy for the treatment of mucositis. However, their use is accompanied by side effects in the gastrointestinal tract. These drawbacks can be avoided by using a local buccal drug delivery which delivers more drugs to the target tissue with less systemic side effects. In this study the selected dosage form for buccal drug delivery is nanofibers due to their high surface area and porosity which facilitated drug release. <This thesis is divided into two chapters: Chapter one: Formulation and in vitro characterization of ketoprofen loaded polymeric electrospun nanofibers. < Nanofibers were successfully prepared with Eudragit S (ES) and Eudragit L (EL) at a concentration 20% (w/v), at a flow rate of 0.5 ml/h and an applied voltage of 15 kV. Two formulations were selected for further evaluations: 20% EL nanofiber loaded with 20% KET (F1) and 20%ES nanofibers loaded with 10% KET (F2). F1 was selected to perform in vivo study due to its high loading capacity reducing the size of nanofiber mat for buccal administration. The complete erosion and drug release from F1 occurred within 2 hours, relieving the pain due to the initial burst release without removing the nanofibers after drug release. <Chapter two: In vivo evaluation of the effect of ketoprofen loaded Eudragit L nanofiber on induced oral mucositis in rabbits. The clinical evaluation of the oral mucositis in rabbits revealed a reduction in mucositis severity in the treated group, while the severity remained the same in the control group. The histopathological study showed ulceration associated with dense inflammatory infiltrates in the control group. The treated group showed an intact epithelium and very mild inflammatory infiltrates. Results confirmed the efficacy of the local buccal delivery of KET loaded EL nanofibers (F1) in treating oral mucositis. |
