Abstract The full length of cDNAs encoding FSHβ, LHβ and common α subunits of gonadotropins of the Egyptian Buffalo were characterized and their sequences were deposited in the GenBank as first record for Egyptian Buffalo. The alignment analysis showed great similarities to Indian buffalo and other ruminants and lower similarities to equines, rodents and primates. Polymorphism in specific loci in beta subunit gene of both FSH and LH was detected using the PCR-SSCP technique followed by sequencing of obtained variants. Three genotypes were found for FSHβ-2 locus, four genotypes for FSHβ-3 locus and seven genotypes for LHβ-2-3 locus. Six out of seven SNPs found in the exon three of FSH were non-synonymous while the SNPs in the other FSH locus and the LHβ-2-3 locus were synonymous. Significant delay in the age of first conception and first calving was found in FF genotype of LHβ-2-3 locus in comparison with FG genotype and significant increase in the calving interval was detected in FF genotype when compared to the CC genotype. Significant decrease in both semen volume and 3 hours post thawing motility was recorded in BB genotype of FSHβ-2 locus when compared to AA genotype with significant decrease in the total sperm abnormalities percent in BB than AA genotype. The CD genotype of FSHβ-3 locus recorded significant increase in sperm concentration/ml than BC genotype but the 3 hours post thawing motility for CD was significantly lower than both AA and BC genotypes. |
