Abstract

The present work was held to appraisal the protective effect of ethanolic extract of garlic and/or fenugreek seed powder against the negative impact of (Cyp) Cyclophosphamide. One hundred and forty rats were split to seven equal groups. First group (control) administrated saline, second group treated with Cyclophosphamide (50mg/kg B.W once every week IM), third group (EEG) administered ethanolic extract of garlic (300 mg/kg B.W orally daily) and fourth group (E.Fsp) treated with ethanolic fenugreek seed powder extract (400 mg/kg B.W orally daily), fifth and sixth groups administrated with ethanolic extract of garlic (300 mg/kg B.W orally daily) and ethanolic fenugreek seed powder extract (400 mg/kg B.W orally daily) respectively and treated with Cyclophosphamide (50mg/kg B.W once every week IM) (EEG+Cyp) and (E.Fsp+Cyp) the seventh group (EEG+E.Fp+Cyp) administered both ethanolic extract of garlic (300 mg/kg B.W orally daily) and fenugreek seed powder (400 mg/kg B.W orally daily) in addition to cyclophosphamide (50mg/kg B.W once every week IM). At 2nd and 4th week whole blood and serum were collected to evaluate hematological, biochemical parameters and immunostimulant parameters also some tissue (liver, kidney and spleen) for histopathology and liver homogenate for determention of oxidative stress and antioxidant markers. The resulted that Cyp treatment induced normocytic normochromic anemia i.e. decrease in erythrocytes count (RBCs), hemoglobin concentration (Hb) and Packed cell volume (PCV %) also decrease in total and different leukocyte counts, moreover the liver enzymes (ALT, AST, ALP and LDH), total bilirubin, direct bilirubin, A/G ratio, triglycerides, cholesterol, glucose, urea and creatinine were increase but total protein, albumin and globulin were decrease, also increase in MDA and decrease in CAT, GSH and SOD moreover induced decrease in all immune parameter (NBT,lysozyme and bactericidal activity) also some cytokine (IL-12, INF-γ) but increased (IL-1β and TNF-α) moreover induced increasement in chromosomal aberration types also decrease the body weight and body weight gain the histopathological showed necrosis and hemorrhage in examined tissues. In contrast the treatment with either ethanolic extract of garlic and/or fenugreek seed powder ameliorated the negative effects of Cyp on hematological, biochemical, oxidative stress, antioxidant markers, immune parameter, structural and numerical chromosomal aberration types, body weight, body weight gain and histopathological results. The conclusion both ethanolic extract of garlic and/or fenugreek seed powder did not have any side effect on body also have protective effect against Cyp. Moreover ethanolic extract of garlic and/or fenugreek seed powder have hepatoprotective, nephroprotective, antimutagenic and immunomodulatory effect.
