Abstract Schotia brachypetala is indigenous to southern Africa. It is considered to have anti-malarial, anti-bacterial, and anti-oxidant activity. It has many components such as tannins, phenolic components, and linolenic acid. We evaluated whether its methanolic leaf extract is of value as an anti-inflammatory agent in two experimental models of inflammation. Rats were divided into six groups in each model of inflammation as follow; normal group, which didn’t receive neither drug nor inflammatory agent, negative control group, which received only inflammatory agent, positive control group which received inflammatory agent and a standard anti-inflammatory drug, and three groups which received an anti-inflammatory agent and Schotia brachypetala methanolic leaf extract in three gradual doses as well as the inflammatory agent. The following parameters were investigated: • Myeloperoxidase activity • Tumor necrosis factor-alpha level • Prostaglandin E2 level. • Paw oedema inhibition • Histopathology The findings of the present study can be summarized as follow: • Schotia brachypetala methanolic leaf extract are considered to be category (5) or unclassified according to OECD guidelines for testing of chemicals. • Schotia brachypetala methanolic leaf extract reduced carrageenin-induced paw oedema in a dose related manner. • Schotia brachypetala methanolic leaf extract, 50 mg/kg, 100 mg/kg and 200 mg/kg dose caused statistically significant reduction of PGE2 content in inflammatory homogenate. • A significant reduction in ear punch weight was found after application of 20mg/ear and 40 mg/ear doses, compared to the group with only croton oil. • Schotia brachypetala methanolic leaf extract reduced MPO activity in the challenged ear tissues in doses 10, 20, and 40 mg/ear compared to the group with only croton oil. • Schotia brachypetala methanolic leaf extract, 10mg/ear, 20mg/ear and 40 mg/ear dose, showed significant inhibition of TNF-α release compared to the group with only croton oil. |
