Abstract

The present work aimed to analyse the direct effect of locally injected BoNT-A on the structure of rat parotid salivary glands including acinar cells, ducts and connective tissue stroma by using light and transmission electron microscope and to analyse morphometrically the change in surface area of acinar cells. Materials and methods: The study was carried out on thirty six adult male wistar rats weighing 200-250 grams. All animals were housed in specially designed wire mesh bottom cages throughout the whole experimental period which lasted for 35 days. The animals were divided into four groups (Nine rats per group). Group1: were euthanized after one week. Group2: were euthanized after two weeks. Group3: were euthanized after four weeks. Group 4: were euthanized after five weeks.
The right parotid glands of each rat were exposed by an incision and injected once with 5U BoNT/A (Botox, allergen, USA) reconstituted in 0.1ml physiologic saline, while the left gland of the same animal were used as internal sham control, injected with 0.1ml physiologic saline only. At 7, 14, 28 and 35 days after operation, the rats were euthanized and both right and left parotid glands were surgically excised, fixed in 10% neutral buffered formalin solution for histological examination using light and electron microscope
