Abstract
In this study we collected 54 blood samples from horse collected from El Zhra farm and 13 tissue samples ( aborted placenta and aborted fetus)from horse , 12 samples from EL Zhra farm and 1 from AHRI . 249 serum samples from horse ,cow, buffalo, sheep, goat and camel from different places in the government as Cairo ,Gezia , Beni Seuf , EL Mina ,Asuiat.
1- 13 tissue samples prepared by passing on BHK21 cell only 1 sample gave cell rounding and detachment from vessels
2. RT PCR used to tested 54 blood samples and 13tissue samples collected from horse tested to detect EHV1,4 and 2 ,all blood samples were negative for EHV 1,4 and 2 ,The tissue samples 1 samples from 13 were positive for EHV1 ,all 13 tissue samples were negative for EHV4 and 2
3- The positive samples for EHV-1 the PCR product were extracted and nucleotide sequence of the purified fragments was determined using an automated DNA sequencer and alignment analysis and phylogenetic analysis of EHV-1 positive samples.
4-AGPT used to tested 249 serum samples to detected EHV-1 and EHV-4 Ab, 44 samples from horse the positive samples 8 as18.18% , 63 samples from donkey the positive samples 12 as 19%,54 samples from cow and buffalo the positive samples 0 as 0% ,24 samples from cow the positive samples 0 as 0% , 8 samples from buffalo the positive samples 0 as0%, 16 samples from sheep the positive samples 0 as 0% ,7 samples from goat the positive samples 0 as 0% , 33 samples from camel the positive samples 0 as 0 %.
5-ELISA used to detected EHV-1 Ab in 107 samples from horse and donkey 44 samples from horse the positive samples 28 samples as 63.6 % ,63 samples from donkey the positive samples 31 samples as 49 % .
6-Compare between the result of AGPT and ELISA 44 samples from horse tested by AGPT and ELISA the positive samples by AGPT 8 samples as 18.18%,The samples tested by ELISA the positive samples 28 samples as 63.6% , 63 samples from donkey tested ,the positive samples by AGPT 12 samples are positive as 19 % ,the samples tested by ELISA 31 samples are positive as 49 % .
