Abstract Anticancer chemotherapy is a successful treatment of tumor to reduce the multiplying rate of tumor cells. Although the immunomodulatory effect of single chemotherapy has been investigated, the effects of combinatorial chemotherapy are not well defined. The aim of this study was to evaluate effects of two of the most common conventional anti-cancer chemotherapeutic drugs: including cyclophosphamide (CTX) and cisplatin (CIS), either individually or in combination on the tumor growth as well as on the total and differential numbers of leukocytes and the phenotypes of myeloid cells. This study includes: groups of adult female Swiss albino (CD1) mice were treated with intraperitoneal (i.p.) injection of 0.5 x 106 cells/mouse Ehrlichs ascites carcinoma (EAC) cell line to form ascetic tumor. After one day of injection, mice were i.p. treated either with PBS, CIS (10μg/mouse), CTX (4mg/mouse) from days 1-4 post tumor challenge or their combination using different sequential treatment protocol of CIS on days 1-4 followed by CTX from days 5-10 or CTX from days 1-4 followed by CIS from days 5-10. Mice were bled on day 11 of tumor challenge for complete blood count (CBC) and then sacrificed to measure: a) total number of EAC, b) total numbers of spleen and bone marrow cells, c) immunophenotypic analysis of myeloid cells, and d) liver function. |
