Abstract

Introduction: Radiation therapy forms an integral component in the management of head and neck cancers. The parotid gland is radiosensitive which leads to Xerostomia and its subsequent harmful effects on patient health. Curcumin extract is one of the most popular natural free radical scavengers and antioxidant; which has ameliorating effect on many organs during radiotherapy. Methods :50 adult male albino rats divided into control group had neither irradiation nor a supplementation. The experimental group was irradiated with a single dose of 15 GY .Sub group : irradiated only Sub group : supplemented with curcumin 200mg/kg orally for 15 days followed by irradiation. Sub group : supplemented with curcumin 200mg/kg orally for 15 day followed by irradiation and the curcumin supplementation extended for two weeks after the irradiation day. Five animals of each experimental subgroup were sacrificed at 1, 7 and 14 days from the irradiation day, the parotid glands were excised and prepared for histological, immunohistochemical, comet assay and malondialdhyde studies. Result: curcumin extract had ameliorating effect on the acinar destruction caused by irradiation; it enhanced acinar cell proliferation, decreased DNA destruction and lipid peroxidation formed as a result of irradiation. Conclusion: Curcumin extract have radio protective effect on irradiated parotid glands.
