Abstract

• This study was done at Medical Biochemistry Department, Faculty of Medicine, Ain Sham University during the period from 2010 – 2012 and included 150 female participants who were admitted to the Obstetrics and Gynecology Hospital, Ain Shams University for delivery during the years.
• The aim of this study was to detect Transforming Growth Factor-β3 and Urotensin II receptor gene expression in placental tissue samples from patients of severe and mild PE as well as normal placental tissue samples from healthy normal women as control and to correlate the data with the different clinical factors and pregnancy outcome to try to find out a role of these genes in the onset and progression of PE.
• The participants were divided into two main groups:
I-Preeclampsia group:
This group was including one hundred preeclampsia cases. Cases that had a history of diabetes, peripheral vascular disease, cardiovascular disease, chronic renal disease, multi-fetal gestation, major fetal abnormalities and other hypertension associated diseases were excluded.
This group was subdivided into two subgroups:
 Severe PE subgroup: fifty severe PE cases (systolic blood pressure ≥ 160/110 mmHg and Proteinuria≥5g/d in 24h urine or ≥+3 by dipstick).
 Mild PE subgroup: fifty mild PE cases (systolic blood pressure≥140/90mmHg and Proteinuria≥300mg/d in 24h urine or ≥+1 by dipstick).
II-Control group: fifty normal primigravida.
• The age of patients in the severe PE subgroup is ranged between18-40 (median=25) years old, in the mild PE subgroup it is ranged between19-40 (median=23) years old and in the control group the range is between19-35 (median=25) years old.
• Full clinical data was obtained from sheets of the patients about systolic and diastolic blood pressure, proteinuria, gestational and maternal age, birth weight, placental weight, pre-pregnancy weight and height of the patient for calculation of the body mass index.
• RNA extraction of all placental samples was done for Semi-quantitative RT-PCR for placental TGF-β3, UT receptor and β-actin as a house keeping gene.
