Abstract

C
hronic myeloid leukemia (CML) is a clonal myeloproliferative disorder originating from multipotent stem cells. Multistep carcinogenesis is referred to the generation of the Ph and BCR-ABL fusion gene expressing p210 with tyrosine kinase activity result in growth promotion, differentiation, resistance to apoptosis, and defect in DNA repair in targeted blood cells
FISH technique permit to realize the precise analysis of chromosomal abnormalities in both metaphase and interphase cells with high sensitivity as it usually analyzes 200 to 500 nuclei. Thus, quantification generated by FISH is more accurate than conventional cytogenetics. FISH detects BCR-ABL in about 95% of CML cases and it detects the approximately 5% of cases with masked translocations that are missed by conventional cytogenetics A new modification of FISH play an important role in detection of del-der 9 which represent an important predictor of disease progression and survival.
In light of this, the present work aimed to detect deletion of derivative chromosome 9 in Ph +ve CML and asses the clinical characteristics of those patients with deletion of derivative chromosome 9 and their clinical outcome
Peripheral blood and /or bone marrow samples were obtained from 25 cases of CML, 20 in chronic phase and 5 in accelerated phase. They were attending the Hematology Oncology Clinic of Ain Shams University Hospital. Their ages ranged from 36 to 45 years, with a median age of 42 years, there were Sixteen (64%) males and nine (36%) females with a male to female ratio of 1.8:1.
