Abstract

MTX is an antimetabolite folate analogue that is widely used either for cancer patients or for patients having autoimmune diseases.This drug competitively inhibits DHFR enzyme and subsequently THF that is essential for DNA and RNA synthesis. Patients undergoing MTX therapy exhibit a variety of side effects affecting several systems. The aim of the present study was to elucidate the changeable histological effect of MTX on some small intestinal mucosal cells and the possible protective role of vitamin A supplementation.Seventy six adult male albino rats, weighting 200 – 250 gms,were used in this study. They were divided into three groups Group” A” (control group Included thirty six rats and was further subdivided into three equal subgroups Subgroup A1 ”negative control”: consisted of twelve rats.Single dose of intraperitoneal saline was given in addition to their standard diet and tap water Subgroup A2”negative control”: consisted of twelve rats.Each rat received 0.5 ml of sunflower oil daily. Subgroup A3”positive control”: consisted of twelve rats.Each rat received vitamin A (5000 I.U/kg/day) once daily by gavage.The rats of this group were sacrificed at the same timing schedule corresponding to that of the treated group.
