Abstract

The aim of the present investigation was to study the possible
beneficial effect of estrogen replacement and/or AT1-RB on
endothelial dysfunction and oxidative stress in ovariectomized rats.
Forty adult female albino rats weighing 150-200 grams, each were
used in this investigation. The rats were divided into the following
groups:
i. Normal (non-ovariectomized) group.
ii. Ovariectomized non-treated group.
iii. Ovariectomized-estradiol treated group.
iv. Ovariectomized angiotensin ΙΙ type Ι receptor blocker treated
group.
v. Ovariectomized with combined estrogen and angiotensin ΙΙ
type Ι receptor blocker treated group.
In each of the previously mentioned groups, serum estradiol, blood
pressure, serum malondialdehyde, serum total antioxidant status,
vascular reactivity of aortic rings to NE and Ang II, and vascular
relaxation of aortic rings to in response Ach and Na+ nitroprusside
were measured.
The results showed that ovariectomy caused significant increase in
systolic blood pressure, serum malondialdehyde and vascular
reactivity to angiotensin II, significant decrease in serum levels of
estradiol, serum total antioxidant status and the vasodilator responses
to acetylcholine, and insignificant change in vascular reactivity to NE
and the vasodilator responses to Na+ nitroprusside when compared to
corresponding values in non-ovariectomized group.
